Dr T, what is your opinion about chemicals that push penetration of topical actives deeper into the skin?

Both hydro- and lipo-philic actives are frequently combined in commercial emulsions. Penetration enhancers in those formulations can include Ethoxydiglycol, Propylene Glycol, Squalane, Dimethyl Isosorbide, alpha bisabolol, etc.

My question is, are the actives wasted if you only have an oil-based penetration enhancer (eg. squalane) or an aqueous based one (eg. DMISO)? Is it the case that say, having squalane in an emulsion can push BOTH hydro- and lipophilic actives deeper?

Penetration depends on complex interaction of many factors including size and chemistry of the actives, surfactants, colloidal effects, use of micelles/liposomes, chemistry of inactives, penetration enhancers, emulsifyers, condition of the skin and so forth. Some actives penetrate adequately w/o any enhancers. Each formula has to be judged separately.

One of the best penetration enhancers is LPO gel, but it is used mainly as a vehicle for transdermal drug delivery and not designed for skin care.
